The
ion-pairing reagent is generally an alkyl sulfate, an alkyl sulfonate, or an
alkylammonium salt. Ion pairing reagents consist of ionic molecules, which have
a charge opposite to component of interest as well as a sufficiently the hydrophobic region, which permits the stationary phase, which interacts with
the associated counter-ions.
- Why do we need an ion-pair HPLC?
The ion-pairing
reagents or ion pair chromatography is used while a sample has ionic molecules
that are very hydrophilic, and therefore reversed-phase retention can be
problematic. It’s also used when other modifications in reversed-phase chromatography (RP-HPLC) conditions did
not succeed to achieve adequate resolution.
